Invitae | RCV001033522 | SCV001196829 | pathogenic | Chuvash polycythemia; Von Hippel-Lindau syndrome | 2022-10-18 | criteria provided, single submitter | clinical testing | This variant is a gross deletion of the genomic region encompassing exon(s) 3 of the VHL gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. A similar copy number variant has been observed in individual(s) with von Hippel-Lindau (VHL) syndrome (PMID: 8069305, 8707293, 19280651). It has also been observed to segregate with disease in related individuals. This variant disrupts the p.Leu188 amino acid residue in VHL. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 7563486, 7987306, 8772572, 11331612, 16452184, 18567581, 23772956). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. |
