This variant is also absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The serine at codon 333 is a highly conserved residue located in the active site of the protein kinase domain (InterPro), and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Additionally, other amino acid substitutions at this codon (Ile, Asn, Gly) have been reported in individuals with HHT and are considered to be causative (Abdalla 2000, Berg 1997, Lux 2013, McDonald 2011, Schulte 2005, see HHT database link). Based on available information, the p.Ser333Arg variant is considered to be likely pathogenic.
