The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline. This variant is not present in population databases (ExAC no frequency). This variant has been reported in individuals affected with complete androgen insensitivity syndrome in the literature (PMID: 8647313), as well as in the Leiden Open-source Variation Database (PMID: 21520333). This variant is also known as p.Leu616Pro in the literature. Experimental studies have shown that this missense change abrogates AR transcriptional activity in vitro (PMID: 8647313, 23106833). In summary, this is a rare missense change that has been reported in affected individuals and has been shown to impair protein function. This evidence indicates that the variant is pathogenic, but additional data is needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
