Four of five in-silico tools predict a benign effect of the variant on protein function. 4/4 computational tools via ALAMUT predict no significant impact on normal splicing although a disruption of an ESE splice site is predicted. In a minigene assay system, this variant was reported to produce 74% of the canonical transcript and low levels of delta 4, delta 4,5 and delta 4,5,6 transcripts (Fraile-Bethencourt_2019). However, the in-vivo impact of these findings have yet to be corroborated by functional studies. The variant allele was found at a frequency of 4e-05 in 251000 control chromosomes. This frequency is not significantly higher than expected for a pathogenic variant in BRCA2 causing Hereditary Breast and Ovarian Cancer (4e-05 vs 0.00075), allowing no conclusion about variant significance. c.352C>T has been reported in the literature in individuals affected with Breast Cancer without strong evidence for causality (Zhang_2016, Bhaskaran_2019). Co-occurrences with other pathogenic variant(s) have been reported (BRCA1 c. 3700_3704del, p. Val1234Glnfs*8; Zografos_2022), providing supporting evidence for a benign role. These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast and Ovarian Cancer. Ten clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
