Labcorp Genetics (formerly Invitae), Labcorp RCV003596846 SCV004293808 pathogenic Congenital factor V deficiency 2023-12-02 criteria provided, single submitter clinical testing This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 218 of the F5 protein (p.Phe218Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive factor V deficiency (PMID: 19052695, 32219828, 34280927). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as 743T>C (F190S). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt F5 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
