Illumina Laboratory Services, Illumina RCV000341877 SCV000352481 uncertain significance Basal laminar drusen 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. It had not been previously curated by ICSL or reported in the Human Gene Mutation Database (HGMD: prior to June 1st, 2018), and was therefore a candidate for classification through an automated scoring system. Utilizing variant allele frequency, disease prevalence and penetrance estimates, and inheritance mode, an automated score was calculated to assess if this variant is too frequent to cause the disease. Based on the score, this variant could not be ruled out of causing disease and therefore its association with disease required further investigation. A literature search was performed for the gene, cDNA change, and amino acid change (if applicable). No publications were found based on this search. This variant was therefore classified as a variant of unknown significance for this disease.
Illumina Laboratory Services, Illumina RCV000408202 SCV000352482 uncertain significance CFH-Related Dense Deposit Disease / Membranoproliferative Glomerulonephritis Type II 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. Illumina Laboratory Services, Illumina RCV000307054 SCV000352483 likely benign Hemolytic uremic syndrome, atypical, susceptibility to, 1 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. Based on the score and internal cut-off values, a variant classified as likely benign is not then subjected to further curation. The score for this variant resulted in a classification of likely benign for this disease.
Illumina Laboratory Services, Illumina RCV000347912 SCV000352484 uncertain significance Age related macular degeneration 4 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. Labcorp Genetics (formerly Invitae), Labcorp RCV001850510 SCV002252421 uncertain significance not provided 2024-12-17 criteria provided, single submitter clinical testing This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 1002 of the CFH protein (p.Gly1002Arg). This variant is present in population databases (rs201816520, gnomAD 0.02%). This missense change has been observed in individual(s) with age-related macular degeneration (PMID: 34508573, 35925583). ClinVar contains an entry for this variant (Variation ID: 294513). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Genome Diagnostics Laboratory, The Hospital for Sick Children RCV002294236 SCV002587566 uncertain significance Atypical hemolytic-uremic syndrome 2022-03-03 criteria provided, single submitter clinical testing
Institute of Human Genetics, Univ. Regensburg, Univ. Regensburg RCV004816522 SCV005072635 uncertain significance Retinal dystrophy 2021-01-01 criteria provided, single submitter clinical testing
Women's Health and Genetics/Laboratory Corporation of America, LabCorp RCV004701396 SCV005205275 uncertain significance not specified 2024-06-26 criteria provided, single submitter clinical testing Variant summary: CFH c.3004G>C (p.Gly1002Arg) results in a non-conservative amino acid change located in the Sushi/SCR/CCP domain (IPR000436) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-05 in 251358 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in CFH causing Age-Related Macular Degeneration, allowing no conclusion about variant significance. c.3004G>C has been reported in the literature in at least one individual affected with Age-Related Macular Degeneration (Farinha_2022). These report(s) do not provide unequivocal conclusions about association of the variant with Age-Related Macular Degeneration. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 35925583). ClinVar contains an entry for this variant (Variation ID: 294513). Based on the evidence outlined above, the variant was classified as uncertain significance.
