This variant has been observed in individuals with suspected familial Mediterranean fever (PMID: 10842288, 23463692, 21246368, 17489852, 16439335). ClinVar contains an entry for this variant (Variation ID: 97480). This variant is not present in population databases (ExAC no frequency). This sequence change replaces methionine with leucine at codon 680 of the MEFV protein (p.Met680Leu). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and leucine.
