The Asn194Ser variant is a conservative amino acid substitution as these residues share similar properties, and are least likely to impact secondary structure. The Asn194 residue is conserved across species. In silico analysis predicts Asn194Ser is damaging to the protein structure/function. Mutations in nearby residues (Asp190Gly, Arg192His, Arg192Cys, Ile195Met, Asp196Asn) have been reported in association with cardiomopathy, further supporting the functional importance of this region of the protein. Furthermore, the Asn194Ser variant was not observed in approximately 6,100 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations.In summary, while Asn194Ser is a good candidate for a disease-causing mutation, with the clinical and molecular information available at this time we cannot unequivocally determine the clinical significance of this variant.
