Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 50790 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.425G>A in individuals affected with Peutz-Jeghers Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. An internal sample reports the variant to co-occur with a likely pathogenic MLH1 variant, c.980_983dupAGCA (p.His329fsX34). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.

This sequence change replaces serine with asparagine at codon 142 of the STK11 protein (p.Ser142Asn). The serine residue is highly conserved and there is a small physicochemical difference between serine and asparagine.
