However, the phenotype of this variant in the presence of other alpha globin variants is unknown. This variant has been reported to have lower HbO levels than expected, suggesting protein instability; however, in vitro studies have shown the protein to be stable (Daud 2011, Molchanova 1994). This variant is only observed on two alleles in the Genome Aggregation Database, indicating it is not a common polymorphism. The glutamic acid at codon 116 is weakly conserved, and computational analyses (SIFT: damaging, PolyPhen-2: benign) predict conflicting effects of this variant on protein structure/function, although these are low confidence predictions. Based on available information, this variant is considered to be likely benign.
