The p.Met1092 residue is not conserved in mammals but one out of five computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) suggest that the c.3275_3276delinsCC variant may impact the protein; however, this information is not predictive enough to assume pathogenicity. This variant is an in-frame deletion-insertion resulting in the replacement of a methionine residue for a threonine residue at codon 1092; the impact of this alteration on PKD1 protein function is not known. The variant occurs outside of the splicing consensus sequence and in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer, HumanSpliceFinder) do not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time.
