The methionine at codon 932 is replaced by valine, an amino acid with highly similar properties. This alteration was not observed in 7,051 unselected female breast cancer patients and was observed with an allele frequency of 0.00009 in 11,241 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

This variant is also reported in ClinVar (Variation ID: 322572), and is only observed on one allele in the Genome Aggregation Database, indicating it is not a common polymorphism. The methionine at codon 932 is moderately conserved, and computational analyses are uncertain whether this variant is neutral or deleterious (REVEL: 0.214). Additional computational analyses (Alamut v.2.11) predict that this variant may impact splicing by creating a novel cryptic acceptor splice site, but without functional studies the effect on splicing is unknown. Due to limited information, the clinical significance of the p.Met932Val variant is uncertain at this time. References: Momozawa Y et al.
