The alanine at codon 1224 is replaced by glycine, an amino acid with similar properties. This variant has been reported in a patient with a blastic plasmacytoid dendritic cell neoplasm (Szczepaniak A et al. Int. J. Hematol., 2019 Jul;110:102-106). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 1224 of the NF1 protein (p.Ala1224Gly).
