The aspartic acid at codon 6390 is replaced by asparagine, an amino acid with highly similar properties. This variant was previously reported in the SNPDatabase as rs370813526. Based on data from the NHLBI Exome Sequencing Project (ESP), the A allele has an overall frequency of approximately 0.01% (1/11952) total alleles studied and 0.01% (1/8242) European American alleles. Based on data from ExAC, the A allele has an overall frequency of approximately 0.01% (12/119826). The highest observed frequency was 0.02% (11/66350) of European (non-Finnish) alleles (Exome Aggregation Consortium (ExAC), Cambridge, MA (URL: http://exac.broadinstitute.org) [Accessed March 3, 2016]). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
