Furthermore, this truncation is located outside of a clinically relevant region of the TTN gene (PMID: 25589632). For these reasons it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a TTN-related disease. This sequence change results in a premature translational stop signal in exon 35 of the TTN mRNA at codon 2740 (p.Trp2740*). While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. |
