Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine RCV000154910 SCV000204592 likely benign not specified 2015-02-19 criteria provided, single submitter clinical testing p.Ile25482Val in exon 275 of TTN: This variant is not expected to have clinical significance due to a lack of conservation across species, including mammals. Of note, >20 species, including many mammals, have a valine (Val) at this position despite high nearby amino acid conservation. In addition, this variant has been identified in 5/11412 Latino chromosomes by the Exome Aggregation Consortium (E xAC, http://exac.broadinstitute.org; dbSNP rs201348580).

Illumina Laboratory Services, Illumina RCV000295690 SCV000421220 uncertain significance Dilated cardiomyopathy 1G 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. It had not been previously curated by ICSL or reported in the Human Gene Mutation Database (HGMD: prior to June 1st, 2018), and was therefore a candidate for classification through an automated scoring system. Utilizing variant allele frequency, disease prevalence and penetrance estimates, and inheritance mode, an automated score was calculated to assess if this variant is too frequent to cause the disease. Based on the score, this variant could not be ruled out of causing disease and therefore its association with disease required further investigation. A literature search was performed for the gene, cDNA change, and amino acid change (if applicable). No publications were found based on this search. This variant was therefore classified as a variant of unknown significance for this disease.
Illumina Laboratory Services, Illumina RCV000348240 SCV000421221 benign Tibial muscular dystrophy 2018-01-13 criteria provided, single submitter clinical testing This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. Based on the score and internal cut-off values, a variant classified as benign is not then subjected to further curation. The score for this variant resulted in a classification of benign for this disease.

Based on the score and internal cut-off values, a variant classified as likely benign is not then subjected to further curation. The score for this variant resulted in a classification of likely benign for this disease.

Women's Health and Genetics/Laboratory Corporation of America, LabCorp RCV000154910 SCV002074545 uncertain significance not specified 2022-01-06 criteria provided, single submitter clinical testing Variant summary: TTN c.76444A>G (p.Ile25482Val) results in a conservative amino acid change located in the A-band region of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00029 in 247830 control chromosomes (gnomAD). c.76444A>G has been reported in the literature in individuals affected with Sudden Unexplained Death/Sudden Infant Death (Campuzano_2015, Campuzano_2018). These reports do not provide unequivocal conclusions about association of the variant with Dilated Cardiomyopathy or Limb-Girdle Muscular Dystrophy, Type 2J. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Five ClinVar submitters have assessed this variant since 2014: two have classified the variant as likely benign, and three as of uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
Ambry Genetics RCV002345503 SCV002648238 likely benign Cardiovascular phenotype 2018-11-28 criteria provided, single submitter clinical testing This alteration is classified as likely benign based on a combination of the following: seen in unaffected individuals, population frequency, intact protein function, lack of segregation with disease, co-occurrence, RNA analysis, in silico models, amino acid conservation, lack of disease association in case-control studies, and/or the mechanism of disease or impacted region is inconsistent with a known cause of pathogenicity.
