RCV004701490 | SCV005202532 | uncertain significance | not specified | 2024-07-12 | criteria provided, single submitter | clinical testing | Variant summary: TTN c.9166C>T (p.Arg3056Cys) results in a non-conservative amino acid change located in the I-band region of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.2e-05 in 250476 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.9166C>T in individuals affected with TTN-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 405039). Based on the evidence outlined above, the variant was classified as uncertain significance. |
