In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 472153). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FLNC protein function with a negative predictive value of 95%. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.

This is a novel variant. There have been no reports of this variant in the literature. The glycine at this residue is highly conserved. In silico predictors to not agree on the potential impact of this missense change. No other variants have been reported in association with disease at this codon and nearby codons. There is no variation at codon 2320 listed in the Genome Aggregation Consortium Dataset (gnomAD; http://gnomad.broadinstitute.org/), which currently includes variant calls on >140,000 unrelated individuals of African, Asian, European, Latino, and Ashkenazi descent. The average coverage at that site in gnomAD is 65x.
