This alteration has been reported in an individual diagnosed with a carotid paraganglioma at age 63 who had no family history of paraganglioma or pheochromocytoma (Bausch B et al. JAMA Oncol, 2017 Sep;3:1204-1212). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
