Labcorp Genetics (formerly Invitae), Labcorp RCV001891357 SCV002162629 uncertain significance Mitochondrial complex II deficiency, nuclear type 1; Paragangliomas 5 2021-08-19 criteria provided, single submitter clinical testing This sequence change replaces methionine with valine at codon 158 of the SDHA protein (p.Met158Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SDHA-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SDHA protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Ambry Genetics RCV003382693 SCV004088713 uncertain significance Hereditary cancer-predisposing syndrome 2023-06-26 criteria provided, single submitter clinical testing The p.M158V variant (also known as c.472A>G), located in coding exon 5 of the SDHA gene, results from an A to G substitution at nucleotide position 472. The methionine at codon 158 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
