Ambry Genetics RCV002424766 SCV002677387 uncertain significance Cardiovascular phenotype 2021-04-07 criteria provided, single submitter clinical testing The p.T2767I variant (also known as c.8300C>T), located in coding exon 24 of the DSP gene, results from a C to T substitution at nucleotide position 8300. The threonine at codon 2767 is replaced by isoleucine, an amino acid with similar properties. This variant was reported in a dilated cardiomyopathy (DCM) cohort; however, clinical details were limited (Mazzarotto F et al. Circulation, 2020 02;141:387-398). This amino acid position is not well conserved in available vertebrate species, and isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
