It was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G470W variant is a semi-D2 conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across mammals but is not conserved in more distantly related species in evolution. A missense mutation in a nearby residue (R468W) has been reported in association with Glut1-DS. In silico analysis predicts this variant is probably damaging to the protein structure/function. However, the possibility that it is a disease-associated mutation cannot be excluded since some individuals with SLC2A1 mutations have been reported to be mildly affected or unaffected due to variability in phenotypic expression and/or reduced penetrance (Mullen et al., 2010; Weber et al., 2008; Suls et al., 2008).
