The p.Leu717Arg variant occurs in alternate transcript NM_007078.2, which corresponds to c.*33568T>G in NM_001080116.1, the primary transcript listed in the Methods.

The leucine at codon 717 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
