This variant, also known as BRCA1 c.2431T>C by alternate nomenclature, has been reported in at least one individual with serous ovarian cancer (Li 2013). BRCA1 Leu771Ser was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Leucine and Serine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. BRCA1 Leu771Ser occurs at a position that is not conserved and is located in the DNA binding domain and a region known to interact with multiple other proteins (Narod 2004, Paul 2004). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, it is unclear whether BRCA1 Leu771Ser is pathogenic or benign. We consider it to be a variant of uncertain significance.
