This variant is also known as 3347delAG and c.3228delAG in the literature. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.

BRCA1 c.3228_3229delAG, also reported as BRCA1 3345delAG and BRCA1 3347delAG using alternate nomenclature, has been observed in several individuals and families with breast and/or ovarian cancer (Malone 1998, Peto 1999, Heimdal 2003, Bj?rge 2004, Gomes 2007, Papi 2009, Seong 2009, H?berg-Vetti 2016) and was shown to segregate with disease in one family (Ottini 2009). We consider this variant to be pathogenic.
