This variant is present in population databases (rs555619169, gnomAD 0.09%). This missense change has been observed in individual(s) with Usher syndrome (PMID: 25404053). ClinVar contains an entry for this variant (Variation ID: 913999). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.

Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00017 in 249952 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in WHRN causing Usher Syndrome (0.00017 vs 0.0038), allowing no conclusion about variant significance. c.2234G>A has been reported in the literature in an individual affected with Usher Syndrome, who also carried other potentially causitive variants (Aparisi_2014). This report does not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 25404053). Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
