Labcorp Genetics (formerly Invitae), Labcorp RCV001313426 SCV001503923 uncertain significance Gorlin syndrome; Medulloblastoma 2024-06-11 criteria provided, single submitter clinical testing This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 397 of the SUFU protein (p.Tyr397Cys). This variant is present in population databases (rs766508656, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with SUFU-related conditions. ClinVar contains an entry for this variant (Variation ID: 1014663). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SUFU protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Ambry Genetics RCV002341631 SCV002641792 uncertain significance Hereditary cancer-predisposing syndrome 2019-09-29 criteria provided, single submitter clinical testing The p.Y397C variant (also known as c.1190A>G), located in coding exon 10 of the SUFU gene, results from an A to G substitution at nucleotide position 1190. The tyrosine at codon 397 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
