RCV001185165 | SCV001351320 | uncertain significance | Cardiomyopathy | 2019-01-07 | criteria provided, single submitter | clinical testing | This variant changes 1 nucleotide in exon 10 of the DSC2 gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Truncations and other loss-of-function variants in the DSC2 gene have been reported in individuals with arrhythmogenic right ventricular cardiomyopathy. However, clinical significance of these variants is not yet fully understood. Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. |
