This variant is present in population databases (rs199473123, ExAC 0.04%). This variant has been reported in an individual affected with Brugada syndrome (PMID: 12639704). ClinVar contains an entry for this variant (Variation ID: 67676). Experimental studies have shown that this missense change does not have a significant effect on sodium channel activity either alone or upon co-expression of wild-type SCN5A (PMID: 24573164). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
